Blood Culture Bottle

Blood culture bottle tubes are for sampling blood for microbiological cultures used for the detection of circulating microorganisms in bacteremia and septicemia. A typical blood culture collection involves drawing blood into two bottles, which together form one “culture” or “set“.

Types of Blood Culture Bottle:

·         Yellow top – pediatric aerobic (0.5–4 mL blood)

·         Green top – adult aerobic (5–10 mL blood)

·         Orange top – anaerobic (5–10 mL blood)

·         Blacktop – mycobacteria (5–10 mL blood)

·         Silver top – mycoplasma (3–5 mL blood)

Contents:

Blood culture bottles contain a growth medium, which encourages microorganisms to multiply, and an anticoagulant that prevents blood from clotting. Sodium polyanethol sulfonate (SPS) is the most commonly used anticoagulant because it does not interfere with the growth of most organisms.

Method of Use:

It is important that the bottles are neither underfilled nor overfilled: underfilling can lead to false negative results as fewer organisms are present in the sample, while overfilling can inhibit microbial growth because the ratio of growth medium to blood is comparatively lower

·         Kindly sterile blood sampling (i.e. hand hygiene, sterile dressing pack, sterile gloves, and clean skin with chlorhexidine)

·         Always take from fresh venepuncture or newly inserted peripheral cannulae and central lines

·         If line sepsis is suspected cultures should be taken from the central line and peripherally

·         blood placed into aerobic and anthe aerobic culture medium

·         remove the dust cap from culture bottles and top swabbed with alcohol (and allowed to dry) before blood is injected

·         maximum yield if the tube is neither under nor over-filled (e.g. 9 mL in adult tubes)

·         fill blood culture tubes before other blood tubes
